This article presents a protocol for evaluating the efficacy and toxicity of RNAs designed to inhibit HIV-1 production, using a reverse transcriptase (RT) assay as a quantitative measure of virus-like particle (VLP) output. The method involves co-transfecting mammalian cells with plasmids and test RNAs, followed by measurement of RT activity in the supernatant to assess the suppression of viral gene expression.
Quantitative reverse transcriptase (RT) assays provide a robust, scalable readout for evaluating the efficacy of RNA-based interventions targeting HIV-1 production. This approach enables early-stage target validation and mechanistic de-risking by linking molecular inhibition to functional suppression of virus-like particle (VLP) output. The method supports portfolio triage and prioritization of candidate gene or drug therapies in antiviral discovery pipelines.
This RT-based assay integrates into the early discovery-to-lead identification continuum, bridging molecular design and preclinical validation for antiviral candidates.
