Barwienie metodą Grama pozwala również na obserwację kształtów bakterii. Okrągłe bakterie nazywane są ziarniakami. Bakterie w kształcie pałeczek nazywane są pałeczkami. A bakterie w kształcie spirali nazywane są spirillą. Co więcej, kolonie bakteryjne można również scharakteryzować gołym okiem, wykorzystując takie cechy, jak kolor, kształt i wielkość kolonii. 
Przyjrzyjmy się teraz grzybom. W przeciwieństwie do bakterii, grzyby są eukariontami, a zatem ich komórki mają organelle i jądro związane błoną. Są one powszechnie klasyfikowane w grupy na podstawie ich strategii rozrodczych. Na przykład Ascomycota, czyli grzyby workowe, do których należą smardze, penicillium i trufle, mają owocniki zwane askokarpami. Smardze są wypełnione tymi strukturami. Jeśli przyjrzymy się im bliżej, zawierają one wiele mniejszych struktur zwanych workami, które z kolei zawierają wiele askospor powstałych podczas rozmnażania płciowego. Jednak workowce często rozmnażają się bezpłciowo przez pączkowanie. Drugą główną grupą grzybów, którą zbadamy, jest Basidiomycota, czyli grzyby klubowe. Większość jadalnych i trujących grzybów w lesie to w rzeczywistości struktury rozrodcze Basidiomycota. W tej grupie to, co nazwalibyśmy kapeluszem grzyba, jest technicznie znane jako bazydiokarp. Trzy domeny życia, bakterie, archeony i eukariota, dzielą życie na trzy odrębne grupy. Podczas gdy wszystkie bakterie należą do domeny Bakterie, grzyby znajdują się w domenie Eukaryota (Grzyby Królestwa). Oprócz archeonów, domena Bakterie obejmują wyłącznie jednokomórkowe prokariota, co oznacza, że ich komórki nie mają zarówno błony jądrowej, jak i organelli związanych z błoną. Natomiast grzyby domeny Eukaryota mają organelle związane z błoną i obejmują zarówno organizmy jednokomórkowe, jak i wielokomórkowe.

Jedną z najczęstszych metod identyfikacji bakterii jest barwienie metodą Grama. Ten test wykorzystuje kolorowe barwniki do określenia składu ściany komórkowej bakterii. Bakterie Gram-dodatnie, które mają grubą ścianę komórkową składającą się z białka zwanego peptydoglikanem, zatrzymują barwnik zwany fioletem krystalicznym i dlatego wydają się fioletowe. Natomiast bakterie Gram-ujemne mają cieńszą ścianę komórkową bez peptydoglikanu i nie zatrzymują fioletu krystalicznego. Zamiast tego komórki te wydają się różowe z powodu barwnika zwanego safraniną. Zastosowanie barwienia metodą Grama często służy jako wstępny krok w identyfikacji gatunku bakterii. Dalsza klasyfikacja wymaga określenia dodatkowych cech. Może to obejmować kształt poszczególnych bakterii, które mogą być ziarniakami (okrągłe), spirillum (spiralne) lub pałeczkami (w kształcie pręcików). Dodatkowo kolor, kształt, połysk, tekstura, a nawet zapach kolonii bakteryjnych mogą być wykorzystane do rozróżnienia gatunków.
<h4>Grzyby</h4>
Grzyby domeny Eukaryota są również bardzo zróżnicowane ekologicznie i mogą mieć różną wielkość, od mikroskopijnych drożdży po największy organizm na ziemi, grzyb miodowy (Armillaria ostoyae). Wszystkie grzyby są heterotroficzne, co oznacza, że muszą pozyskiwać pożywienie ze środowiska. Organizmy te wykazują również różnorodność w swoich specyficznych działaniach, niszach i potencjalnym wpływie na zdrowie człowieka. Na przykład niektóre grzyby są spożywane lub wykorzystywane do produkcji żywności przez ludzi, podczas gdy inne mogą powodować ciężkie, a nawet śmiertelne infekcje2.
W obrębie królestwa grzybów istnieją dwa odrębne podziały taksonomiczne oparte na strategii reprodukcyjnej gatunku. Po pierwsze, Ascomycota, znane jako grzyby workowe, mają strukturę rozrodczą zwaną workiem. Razem wiele worków tworzy askokarp, czyli "owocnik" grzybów. Grzyby te mogą rozmnażać się płciowo lub bezpłciowo przez pączkowanie. <h4>Znaczenie mikrobiologii</h4>
Ogólnie rzecz biorąc, oczywiste jest, że różnorodne funkcje i właściwości mikroorganizmów sprawiają, że ich identyfikacja jest niezwykle ważna dla zdrowia i dobrego samopoczucia człowieka. Chociaż do tej pory zidentyfikowano ogromną liczbę gatunków grzybów i bakterii, szacuje się, że miliony innych nie zostały jeszcze zidentyfikowane i sklasyfikowane. Ponadto ludzkie dyscypliny, które badają grzyby i bakterie, są tak różnorodne, jak same organizmy i bez wątpienia będą się nadal rozwijać. Znaczenie mikroorganizmów w medycynie jest obecnie bardziej powszechne niż kiedykolwiek, ponieważ wielooporne na antybiotyki gatunki bakterii zakaźnych zagrażają zdrowiu ludzkiemu. Zrozumienie struktury i cech biologicznych gatunków zakaźnych i niezakaźnych dostarcza kluczowych informacji niezbędnych do tworzenia nowych terapii antybiotykowych i strategii zapobiegawczych3-4.
Mikrobiologia ma również kluczowe znaczenie dla przemysłu spożywczego. Wiele produktów, w tym jogurt, ser, kombucha, chleb i alkohol, jest wytwarzanych przy użyciu określonych kategorii lub gatunków drobnoustrojów. how scientists identify bacteria and fungi

Bacteria and fungi are highly diverse organisms, and scientists identify them by looking at their structure, growth, and role in health. Some species help people, while others can cause disease, so learning how to tell them apart matters in biology and medicine.

Biological taxonomists group organisms by evolutionary relatedness. Life is divided into three domains: Bacteria, Archaea, and Eukaryota. Bacteria belong to the domain Bacteria, which includes single-celled prokaryotes. Prokaryotic cells lack a nuclear membrane and membrane-bound organelles. Fungi belong to the domain Eukaryota, in Kingdom Fungi. They have membrane-bound organelles and can be either unicellular or multicellular.

Bacteria live almost everywhere on Earth, and their diversity is very large. This group includes helpful species such as Streptococcus thermophilus, which is used to make fermented foods like yogurt. It also includes harmful species such as Bacillus anthracis, whose spores cause anthrax. Microbiologists use several tests to identify bacteria. One common test is Gram staining, which uses colored dyes to show cell wall structure.

Gram-positive bacteria have a thick cell wall made of peptidoglycan, so they retain crystal violet and appear purple. Gram-negative bacteria have a thinner cell wall without peptidoglycan, so they do not keep crystal violet. Instead, they take up safranin and appear pink. Gram staining is often a first step in bacterial identification. Scientists then look at other traits, including cell shape. Cocci are round, spirillum are spiral, and bacilli are rod-shaped. Colony color, shape, sheen, texture, and even smell can also help distinguish species.

Fungi are also diverse and can range from microscopic yeast to the honey fungus, Armillaria ostoyae, which is the largest organism on Earth. Fungi are heterotrophic, which means they must get food from their environment. Some fungi are used in food production or are eaten by people, while others can cause severe or even deadly infections.

Within Kingdom Fungi, two major divisions are based on reproduction. Ascomycota are the sac fungi. They have a reproductive structure called an ascus, and many asci form an ascocarp, or fruiting body. These fungi can reproduce sexually or asexually by budding. This group includes yeast used in brewing beer and baking bread, as well as edible fungi like morels and truffles.

Basidiomycota are commonly called club fungi because they have a club-shaped basidium. Most Basidiomycota reproduce sexually. Two haploid mycelia join to form a basidiocarp, which is the cap of a button mushroom. On the underside, gills lined with basidia produce haploid nuclei that later form basidiospores. Wind spreads these spores. This group includes many button mushrooms and wild puff mushrooms.

Studying microbes is important for human health and for food production. Millions of species may still need to be identified and classified, and microbiology continues to grow as a field. This work is especially important now because multi-antibiotic-resistant bacteria can threaten human health. Knowing microbial structure and biology helps scientists develop new antibiotics and prevention strategies. Microbiology also supports the food industry, where microbes are used to make yogurt, cheese, kombucha, bread, and alcohol, while other compounds and preservatives are designed to limit unwanted microbial growth.
