Ribozymes

Rybozymy to specjalne rodzaje RNA, które mogą działać jako enzymy. 
Substratami dla większości naturalnie występujących rybozymów są wiązania fosfodiestrowe RNA. Jedynym znanym wyjątkiem od tej reguły jest rybosomalny RNA 23S w rybosomie bakteryjnym, który katalizuje tworzenie wiązań peptydowych.
Rybozymy zostały znalezione we wszystkich typach organizmów i są podobne do enzymów białkowych, ponieważ oba zwiększają szybkość reakcji. Wiele rybozymów potrzebuje jonów metali, takich jak magnez, jako kofaktory do katalizowania reakcji.
Niektóre klasy intronów mogą działać jako rybozymy. Introny są podzielone na pięć odrębnych klas – jądrowe mRNA, jądrowe tRNA, archeony, introny grupy pierwszej i grupy drugiej.
Introny grupy pierwszej i drugiej to duże rybozymy o długości kilkuset nukleotydów, które znajdują się w mitochondriach grzybowych i roślinnych, chloroplastach, bakteriofagach i wirusach eukariotycznych. 
Mogą one samodzielnie łączyć się bez pomocy jakichkolwiek białek, podczas gdy introny jądrowe są splicowane przez spliceosom, kompleks enzymatyczny zawierający RNA i białka. 
Małe rybozymy mają zwykle długość od 50 do 150 nukleotydów i są samorozszczepiającymi się motywami sekwencji nukleotydów. Można je znaleźć w wielu wirusach roślinnych RNA, a także w wirusie zapalenia wątroby typu delta, ludzkim patogenie. 
Proces replikacji w tych wirusach wytwarza długie RNA niosące wiele jednostek genomu wirusa, z których każda przenosi mały rybozymy, takie jak młot. Te regularnie rozmieszczone enzymy młota ulegają samodzielnemu rozszczepieniu, co powoduje rozpad długiego RNA na poszczególne segmenty genomu.
Większość rybozymów rozszczepia własną sekwencję nukleotydową, jednak rybonukleaza P może rozszczepiać inne cząsteczki RNA. Rybonukleaza P znajduje się w niektórych bakteriach i przetwarza prekursor tRNA w celu wytworzenia dojrzałego końca 5'.
Wiadomo, że naturalnie dostępne rybozymy katalizują wąski zakres reakcji, takich jak transfer fosforylu w kwasach nukleinowych i tworzenie wiązań peptydowych w białkach. Naukowcy zsyntetyzowali w laboratorium sztuczne rybozymy, które mogą przeprowadzać szeroki zakres reakcji, takich jak tworzenie wiązań węgiel-węgiel i reakcje utleniania-redukcji.

Termin rybozym jest używany w odniesieniu do RNA, które może działać jako enzym. Rybozymy występują głównie w wybranych wirusach, bakteriach, organellach roślinnych i niższych eukariontach. Rybozymy odkryto po raz pierwszy w 1982 r., kiedy w laboratorium Toma Cecha zaobserwowano introny grupy I działające jako enzymy. Wkrótce potem laboratorium Sida Altmana odkryło inny rybozym, rybonulceazę P. Zarówno Cech, jak i Altman otrzymali w 1989 roku Nagrodę Nobla w dziedzinie chemii za prace nad rybozymami. Rybozymy można podzielić na dwie grupy w zależności od ich wielkości – duże i małe. Duże rybozymy mogą mieć różną wielkość od kilkuset do kilku tysięcy nukleotydów. Introny typu I i II oraz bakteryjna rybonukleaza P to duże rybozymy. Małe rybozymy mają długość od 30 do 150 nukleotydów. Występują w wielu chorobotwórczych wirusach roślinnych oraz w wirusie zapalenia wątroby typu delta (HDV), ludzkim patogenie. Hammerhead, hairpin, HDV i Varkud to powszechne typy małych rybozymów. Większość dużych rybozymów potrzebuje do swojej aktywności jonów metali, zwłaszcza Mg2+, ale jony metali nie są potrzebne większości małych rybozymów. Rybozym glmS w mRNA glmS jest unikalnym a-rybozymem, ponieważ działa również jako rybozym, gdy 6-fosforan glukozaminy jest obecny w wysokich stężeniach. Większość naturalnie występujących rybozymów katalizuje samorozszczepiające się wiązania fosfodiestrowe obecne w ich własnym RNA. Ribozymes are RNA molecules that can act as enzymes. They help speed up specific chemical reactions without being protein enzymes. This makes them an important example of RNA with more than one job in the cell.

Some ribozymes can cut RNA molecules, and others can join them together. This shows that RNA can do catalytic work as well as carry genetic information. Ribozymes are a key part of how scientists think about the many roles of RNA in biology.
