Zazwyczaj drugorzędowe N-nitrozoaminy mają mniejsze znaczenie w zastosowaniu komercyjnym. Są one jednak badane ze względu na ich silne właściwości rakotwórcze. W Norwegii (1962) wybuch zatrucia pokarmowego u owiec przypisano mączce rybnej poddanej działaniu azotynów. Azotyn sodu służy do konserwowania różnego rodzaju żywności i mięsa. W żołądku reagują z kwasem żołądkowym, tworząc kwas azotawy. Kwas azotawy reaguje następnie z aminami drugorzędowymi zawartymi w żywności, dając wysoce rakotwórcze N-nitrozoaminy. Z tego powodu użycie azotynu sodu zostało ograniczone przez FDA do 50 do 125 ppm w konserwowaniu mięsa. N-nitrozoaminy występują także w papierosach i dymie tytoniowym.

nitrosamine formation from secondary amines

Secondary amines can form N-nitrosamines when they react with nitrous acid. Nitrous acid is weak and unstable, so it is made in the reaction mixture from sodium nitrite and a strong acid such as hydrochloric acid or sulfuric acid. The reaction is carried out in cold conditions.

In acid, nitrous acid becomes protonated. It then loses water and forms the nitrosonium ion, a reactive electrophile. The secondary amine acts as a nucleophile and attacks this ion. That step gives an N-nitrosammonium ion.

Water then removes a proton from the ion to make the secondary N-nitrosamine. These products can be separated from the mixture as oily yellow liquids. Although they are not usually important for commercial use, they are studied because many are potent carcinogens.

The transcript also links this chemistry to food safety. In 1962, a food-poisoning outbreak in sheep in Norway was traced to nitrite-treated fish meal. Sodium nitrite is used to preserve foods and meat, but in the stomach it can form nitrous acid. That acid can react with secondary amines in food to produce highly carcinogenic N-nitrosamines, so FDA limits sodium nitrite in meat preservation to 50 to 125 ppm. N-nitrosamines are also found in cigarette and tobacco smoke.
