Liczba obrotów enzymu to maksymalna liczba cząsteczek substratu, które może on przekształcić w jednostce czasu. Liczba obrotów dla większości enzymów waha się od 1 do 1000 cząsteczek na sekundę. Katalaza ma największą znaną liczbę obrotów, zdolną do przekształcenia do 2,8 × 106 cząsteczek nadtlenku wodoru w wodę i tlen na sekundę. Lizozym ma najniższą znaną liczbę obrotu wynoszącą pół cząsteczki na sekundę. Chymotrypsyna jest enzymem trzustkowym rozkładającym białka podczas trawienia. Liczba obrotów chymotrypsyny wynosi 100 cząsteczek na sekundę. Gdyby ta reakcja zachodziła bez katalizatora, rozerwanie wiązań peptydowych w wodzie o neutralnym pH zajęłoby setki lat. Zatem wysoka liczba obrotów chymotrypsyny pomaga w szybkim trawieniu białek w jelicie. Enzym oksygenaza karboksylazy rybulozo-1,5-bisfosforanu, czyli RuBisCO, ma bardzo niską liczbę obrotów, wiążąc 3 cząsteczki CO2 na sekundę i jest jednym z najwolniejszych enzymów. Jednak obfitość RuBisCO w przyrodzie rekompensuje niski obrót. RuBisCO stanowi około 50% całkowitego białka występującego w liściach. Enzym o wysokim współczynniku obrotów niekoniecznie musi być wysoce wydajny. Wydajność katalityczną enzymu wyraża się stosunkiem liczby obrotów, kcat, do powinowactwa, KM. Innymi słowy, aby enzym był skuteczny, powinien mieć również niski KM substratu. Średnia wydajność katalityczna większości enzymów wynosi około 105 M-1s-1, co oznacza, że są one umiarkowanie wydajne. Niewiele enzymów o wydajności katalitycznej pomiędzy 108-109 M-1s-1 jest superwydajnych lub doskonałych katalitycznie.

Enzymes can work at very different speeds, and their turnover number shows how fast they convert substrate molecules into products. Turnover number is the maximum number of substrate molecules an enzyme can transform per unit time. For most enzymes, this value is between 1 and 1000 molecules per second.

Some enzymes are especially fast. Catalase has the highest known turnover number and can convert up to 2.8×10^6 molecules of hydrogen peroxide into water and oxygen each second. Lysozyme has the lowest known turnover number, at half a molecule per second.

Digestion also depends on enzyme speed. Chymotrypsin is a pancreatic enzyme that breaks down proteins during digestion, and its turnover number is 100 molecules per second. Without an enzyme, peptide bonds would take hundreds of years to break in water at neutral pH. Chymotrypsin helps make protein digestion in the intestine much faster.

Not every slow enzyme is unimportant. RuBisCO, or ribulose 1,5-bisphosphate carboxylase oxygenase, has a very low turnover number and fixes only 3 molecules of CO2 per second. It is one of the slowest enzymes, but it is very abundant in nature. RuBisCO makes up around 50% of the total protein found in leaves.

A high turnover number does not always mean high catalytic efficiency. Catalytic efficiency is the ratio of turnover number, kcat, to affinity, KM. An enzyme also needs a low KM for its substrate to be efficient. The average catalytic efficiency of most enzymes is about 10^5 M^-1 s^-1, so they are moderately efficient. A few enzymes reach 10^8 to 10^9 M^-1 s^-1 and are called superefficient or catalytically perfect.
