Delipidation Relipidation

Delipidation-relipidation is a biochemical strategy that removes lipids associated with a protein or membrane preparation and then restores selected lipids to examine how they influence structure and function. Delipidation commonly uses controlled extraction or detergent treatment, while relipidation introduces defined lipid compositions under conditions that support reassociation or membrane reconstitution. Comparing untreated, delipidated, and relipidated samples can reveal lipid requirements for protein stability, activity, folding, and interactions with membranes. In biochemistry, this approach helps distinguish intrinsic protein properties from effects caused by the surrounding lipid environment and supports the design of model membranes and functional reconstitution systems.

A Whole-Mount Organ Staining Technique for Three-Dimensional Imaging of the Kidney

2025

This video demonstrates an assay for the whole-mount staining of murine kidneys using a tissue-clearing method called CUBIC (clear, unobstructed brain/body imaging cocktails and computational analysis) and immunofluorescent staining. The kidney is subjected to tissue clearing, wherein the kidney is delipidated, decolorized, and treated with a refractive index matching solution to make the tissue transparent for optimum three-dimensional imaging. The tissue is stained with fluorescently labeled...
