Deuterium Uptake Measurement

Deuterium uptake measurement is a biophysical approach for tracking how hydrogen atoms in a biomolecule are replaced by deuterium, providing information about molecular structure, flexibility, and solvent exposure. In hydrogen-deuterium exchange experiments, proteins are exposed to deuterated water, allowing accessible backbone hydrogens to exchange according to local environment; mass spectrometry then detects the resulting mass increase and its change over time. In biochemistry, this method helps map protein folding, conformational dynamics, ligand or protein interactions, and structural changes that may not be apparent from static structures, supporting mechanistic studies of biomolecular function and regulation.
