Leukocyte Lipid Profile

Leukocyte lipid profile is the composition and relative abundance of lipids in white blood cells, providing a biochemical view of membrane structure, energy storage, and cell signaling. Researchers typically isolate leukocytes, extract their lipids, and separate and quantify classes such as phospholipids, cholesterol, triglycerides, and signaling lipids using chromatographic or mass spectrometric methods. Because immune activation can remodel lipid synthesis, storage, and turnover, profiling helps connect leukocyte metabolism with inflammation and cellular function. In biochemistry, this approach supports studies of immune responses, metabolic disorders, inflammatory disease, and potential lipid-based biomarkers of physiological or pathological states.

A rapid method to obtain infiltrating leukocytes from the murine brain is described. This method utilizes a continuous Percoll gradient and discontinuous Ficoll gradient to select and purify the leukocyte-enriched layer. Isolated leukocytes may then be characterized by flow cytometric measurements.

Mammalian integument contains solvent-extractable lipids that can provide chemical compositions characteristic of individual species. This paper presents a routine method for separating broad lipid classes isolated from integumentary tissues using thin layer chromatography and determining the triacylglyceride profile by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry.

Selective Harvesting of Marginating-hepatic Leukocytes

Marginating-hepatic leukocytes exhibit unique characteristics and distinct immunological functions compared to other leukocyte populations. Here we describe a method for selective harvesting of this specific hepatic cell population, through forced perfusion of the liver of rats or mice. Marginating-hepatic leukocytes seem critical in determining susceptibility to hepatic-related diseases and metastases.
