Rna Structure Stability

RNA structure stability is the ability of an RNA molecule to maintain functional three-dimensional conformations under particular chemical and physical conditions, a property that influences folding, activity, and persistence. It arises from intramolecular base pairing and base stacking that form secondary structures, together with tertiary interactions and interactions with ions, especially magnesium, that help compact and stabilize the molecule; temperature, pH, and ribonucleases can shift this balance. In biochemistry, analyzing RNA stability helps explain catalytic RNAs, riboswitches, and gene-regulatory elements, while guiding RNA design, purification, storage, and the development of RNA-based research and therapeutic strategies.

Biophysical and biochemical studies of interactions among membrane-embedded protein domains face many technical challenges, the first of which is obtaining appropriate study material. This article describes a protocol for producing and purifying disulfide-stabilized transmembrane peptide complexes that are suitable for structural analysis by solution nuclear magnetic resonance (NMR) and other analytical applications.
