Dna Extraction Analysis

DNA extraction analysis is the assessment of DNA isolated from biological samples to determine its quantity, purity, and suitability for downstream research. The process typically begins with cell lysis, followed by removal of proteins and other contaminants; DNA is then recovered through alcohol precipitation or binding to a silica-based column and evaluated by spectrophotometry or fluorescence. Measurements such as concentration and absorbance ratios indicate whether the extract is intact and free from inhibitors. In bioengineering, reliable DNA extraction analysis supports polymerase chain reaction, sequencing, cloning, genetic modification, and other workflows that depend on high-quality nucleic acid templates.

Here we present a new approach to identify plant viruses with double-strand DNA genomes. We use standard methods to extract DNA and RNA from infected leaves and carry out next-generation sequencing. Bioinformatic tools assemble sequences into contigs, identify contigs representing virus genomes and assign genomes to taxonomic groups.

Reliable and accurate forms of field-deployable DNA analysis are crucial for many industries. This paper offers two methods for environmental microbial analysis in the field: 1) the isolation of microbial DNA from environmental samples using a biological extraction field kit, and 2) the analysis of DNA via the field qPCR unit.

Extraction of High Molecular Weight DNA from Microbial Mats

We provide an improved protocol for extracting high molecular weight DNA from hypersaline microbial mats. Microbial cells are separated from the mat matrix prior to DNA extraction and purification. This enhances the concentrations, quality, and size of the DNA. The protocol may be used for other refractory samples.

Xenopus egg extract is a useful model system to investigate the DNA damage checkpoint. This protocol is for the preparation of Xenopus egg extracts and DNA damage checkpoint inducing reagents. These techniques are adaptable to a variety of DNA damaging approaches in the study of the DNA damage checkpoint signaling.
