Optical Assessment

Optical assessment is the use of light-based measurements to characterize the structure, composition, or function of biological materials and engineered systems. It works by analyzing how a sample absorbs, scatters, transmits, or emits light, often through imaging, spectroscopy, or fluorescence measurements that convert optical signals into quantitative data. In bioengineering, these methods support noninvasive monitoring of cells and tissues, evaluation of biomaterials, and characterization of engineered constructs. Because optical measurements can provide spatial and functional information without extensive sample preparation, they help researchers track biological changes, assess performance, and refine diagnostic, therapeutic, and tissue-engineering technologies.

Optical-resolution photoacoustic microscopy (OR-PAM) is an emerging technology capable of imaging optical absorption contrasts in vivo with cellular resolution and sensitivity. We describe the adaptation of optical projection tomography (OPT)1 to imaging in the near infrared spectrum, and the implementation of a number of computational tools. These protocols enable assessments of pancreatic β-cell mass (BCM) in larger specimens, increase the multichannel capacity of the technique and increase the quality of OPT data.
