Cetylpyridinium Chloride Extraction

Cetylpyridinium chloride extraction is a biological sample-preparation method that uses a cationic surfactant to separate nucleic acids or other negatively charged biomolecules from complex mixtures. Under controlled conditions, cetylpyridinium ions interact electrostatically with acidic cellular components, forming insoluble complexes that can be collected by centrifugation, washed, and redissolved for analysis. This approach can concentrate target material while reducing proteins, lipids, polysaccharides, and other substances that interfere with downstream assays. In biological techniques, the resulting extracts support applications such as nucleic acid detection, amplification, characterization, and comparative analysis of samples.

We describe a method for extraction of high molecular weight genomic DNA from planktonic biomass concentrated on 0.22 μm Sterivex filters, followed by cesium chloride density gradient centrifugation for purification.

In this video, we describe a method for the detection of osteoblast-secreted calcium deposits using an anionic dye, Alizarin Red S. The cultured osteoblasts are first stained and then the dye is recovered for colorimetric quantification of calcium deposits or the degree of osteoblast mineralization.

Dynamic Electrochemical Measurement of Chloride Ions

Dynamic measurement of chloride ions is presented. Transition time of an Ag/AgCl electrode, during a chronopotentiometric technique, can give the concentration of chloride ions in electrolyte. This method does not require a stable conventional reference electrode.
