Mosquito Insecticide Test

A mosquito insecticide test is a biological assay used to measure how effectively an insecticide affects mosquitoes and to identify reduced susceptibility in a population. In a controlled procedure, mosquitoes encounter a defined insecticide dose for a specified period, after which researchers record knockdown, mortality, or recovery under standardized conditions. These results help distinguish susceptible mosquitoes from resistant ones and support surveillance of insecticide resistance. The test informs the selection and evaluation of compounds used in mosquito-control programs, while providing a practical foundation for studying insecticide action and improving strategies to reduce vector-borne disease transmission.

Topical Testing Assay: A Technique to Evaluate the Insecticidal Potential of Dopamine Receptor Antagonists on Adult Female Mosquitoes by Topical Application

2025

In this video, we demonstrate the topical testing assay to determine the toxicity of a potential insecticide on adult female mosquitoes. Using a dopamine receptor antagonist that causes lethal inhibition of neurotransmission in mosquitoes, this method describes a way to evaluate the toxic effects of insecticides upon topical application.

Protocols are described for assessing the toxicity of chemistries to immature and adult mosquitoes for development as new classes of larvicides, adulticides and endectocides. The protocols enable high-throughput testing of multiple chemistries at single-point dose and subsequent evaluation via dose response assay to determine toxicity on contact or via ingestion.

We describe the methodology and importance of the topical application bioassay to measure insecticide susceptibility in mosquitoes and fruit flies. The presented assay is high-throughput, utilizes insect mass-thus allowing for calculating a mass-relativized lethal dose instead of concentration-and likely has lower variability than other similar methods.
