Iodide-yfp-gjic Assay

The iodide-YFP-GJIC assay is a fluorescence-based method for measuring gap junction intercellular communication, the direct transfer of small molecules between neighboring cells. Cells expressing an iodide-sensitive yellow fluorescent protein are exposed to iodide, which enters communicating cells through gap junction channels and quenches YFP fluorescence; the rate and extent of fluorescence loss indicate junctional transfer. In biology research, this assay provides a quantitative readout of gap junction activity and helps evaluate connexin function, cellular connectivity, and changes in communication caused by experimental conditions or candidate treatments.

An accurate method for the assessment of cell death is described. The protocol improves upon conventional Annexin V/ propidium iodide (PI) protocols, which display up to 40% false- positive events in cell lines and primary cells from a broad range of animal models.

Multiphoton microscopy of whole mouse organs is possible by optically clearing the organ before imaging, but not all protocols preserve the fluorescent signal of fluorescent proteins. Using an optical clearing method with ethanol-based dehydration and benzyl alcohol:benzyl benzoate clearing, we show high-resolution multiphoton images of whole mouse brain expressing YFP.
