Ion Channel Expression

Ion channel expression is the production, processing, and delivery of membrane proteins that control the movement of ions across cell membranes. It begins when genes encoding channel subunits are transcribed and translated, followed by protein folding, assembly, modification, and trafficking to the plasma membrane or intracellular compartments, where channels open or close in response to voltage, ligands, or mechanical signals. In biology, measuring and manipulating ion channel expression helps explain electrical signaling, muscle contraction, secretion, and cellular homeostasis. These studies also reveal how altered channel abundance or localization contributes to disease and support the development of targeted therapies.

Afferent sensory neurons signal sensory information from the periphery to the central nervous system. Identifying specific afferent neurons will help in understanding their physiology. We describe a method of retrograde labeling to identify afferent neurons, and study the voltage-gated ion channels in these neurons using patch clamp electrophysiology and immunocytochemistry.

The purification of ion channels is often challenging, but once achieved, it can potentially allow in vitro investigations of the functions and structures of the channels. Here, we describe the stepwise procedures for the expression and purification of mammalian bestrophin proteins, a family of Ca2+-activated Cl- channels.
