Ligation-mediated Pcr

Ligation-mediated PCR (LM-PCR) is a molecular technique that amplifies DNA fragments after attaching known adapter sequences to their ends, enabling analysis of otherwise undefined or difficult-to-amplify regions. The method uses DNA ligase to join adapters to compatible fragment ends, followed by PCR with an adapter-specific primer and a primer targeting the sequence of interest; repeated thermal cycling exponentially increases selected products. In biology, LM-PCR supports genomic mapping, detection of DNA breaks, analysis of restriction fragments, and preparation of DNA for sequencing. By linking unknown DNA ends to a known sequence, it provides a practical route for identifying fragment boundaries and characterizing complex genomic samples.
