Oligonucleotide Annealing

Oligonucleotide annealing is the process by which complementary single-stranded DNA or RNA molecules align and form a stable, double-stranded structure through hydrogen bonding between matching bases. The reaction depends on sequence complementarity and conditions such as temperature, salt concentration, and oligonucleotide concentration, which influence hybridization and duplex stability. In biology, annealed oligonucleotides support molecular methods including polymerase chain reaction, DNA sequencing, cloning, hybridization assays, and gene regulation studies. Controlling annealing conditions improves probe binding, primer specificity, and experimental reproducibility, making this process essential for accurate nucleic acid detection and manipulation.
