Metal-depleted Medium

Metal-depleted medium is a laboratory growth medium formulated with greatly reduced concentrations of metal ions, allowing researchers to study how microorganisms or cultured cells respond to metal limitation. It is prepared by omitting metal supplements or removing residual ions, often through chelating agents that bind metals and lower their bioavailable fraction; careful control helps prevent contamination from reagents and vessels. In biology, this medium supports experiments on metal homeostasis, transport, enzyme function, stress responses, and nutrient requirements. Comparing growth, gene expression, or metabolic activity with metal-replete controls can reveal how organisms acquire and use essential metals and respond to deficiency.

Here we present a protocol to synthesize novel, high-aspect ratio biocomposites under biological conditions and in liquid media. The biocomposites scale from nanometers to micrometers in diameter and length, respectively. Copper nanoparticles (CNPs) and copper sulfate combined with cystine are the key components for the synthesis.

Macrophages play a central role in homeostasis and pathology in many tissues. The protocol presented here describes methods for depleting macrophages in vivo, deriving polarized macrophages from bone marrow aspirates, and adoptively transferring macrophages into mice. These techniques allow determination of the role that polarized macrophages play in health and disease.
