Silver Grain Detection

Silver grain detection is an autoradiographic method that visualizes radioactive molecules or cellular structures by locating metallic silver grains in a photographic emulsion. In biological samples, radiation emitted by an incorporated or bound radioisotope interacts with nearby silver halide crystals, reducing them during development and producing dark grains that mark the tracer’s position. Researchers examine grain distribution with light or electron microscopy to identify labeled cells, organelles, nucleic acids, or sites of molecular activity. The method supports studies of DNA and RNA synthesis, cell division, receptor localization, and tissue organization, while grain density can provide a relative measure of radioactive signal.

This report describes techniques to isolate and purify sulfated glycosaminoglycans (GAGs) from biological samples and a polyacrylamide gel electrophoresis approach to approximate their size. GAGs contribute to tissue structure and influence signaling processes via electrostatic interaction with proteins. GAG polymer length contributes to their binding affinity for cognate ligands.

Femtosecond-laser direct-writing is frequently used to create three-dimensional (3D) patterns in polymers and glasses. However, patterning metals in 3D remains a challenge. We describe a method for fabricating silver nanostructures embedded inside a polymer matrix using a femtosecond laser centered at 800 nm.
