Cytokine Therapy

Cytokine therapy is a treatment approach that uses immune-signaling proteins to regulate host defense, inflammation, and cell growth, with important applications in cancer research. Cytokines such as interferons and interleukins bind specific receptors on immune or tumor cells, activating intracellular signaling pathways that can stimulate cytotoxic lymphocytes, alter cell proliferation, or promote tumor-cell death. In oncology, these agents may enhance antitumor immunity directly or support other immunotherapies, while their effects depend on dose, delivery, tumor context, and interactions within the tumor microenvironment. Studying cytokine therapy helps researchers improve treatment selectivity, manage inflammatory toxicities, and develop more effective combination strategies.

Described here is a method for producing exogenous cytokine in patient-derived xenograft (PDX) mice via weekly intraperitoneal injection of a cytokine-transduced stromal cell line. This method broadens the utility of PDX and provides the option for transient or sustained exogenous cytokine delivery in a multitude of PDX models.
