Omentum Dissection

Omentum dissection is the controlled separation and removal of the omentum, a fatty fold of peritoneal tissue that can participate in abdominal tumor spread and immune responses. The procedure uses careful sharp and blunt dissection along anatomical tissue planes, with attention to nearby organs and blood vessels, to isolate the omental specimen while preserving relevant structures. In cancer research, dissected omentum supports analysis of metastatic implantation, tumor–stroma interactions, immune-cell composition, and cancer-associated molecular changes. Standardized specimen collection also enables comparison across disease models and patient samples, improving understanding of peritoneal metastasis and informing surgical, diagnostic, and therapeutic strategies.

In this video, we establish an ex vivo omental organ culture overlaid with GFP-expressing ovarian cancer cells to study ovarian cancer colonization within the omental explant. This protocol analyzes immune cell structures within the omentum known as milky spots that promote metastases.

Mesothelial Cell Isolation: A Technique to Isolate Primary Mesothelial Cells From Human Omentum Tissue Specimen

2023

This video describes the protocol to isolate primary human mesothelial cells from the human omentum specimen. The mesothelial cells can be used to establish an in vitro ovarian cancer model to investigate ovarian cancer cell adhesion, invasion, and proliferation.
