Penicillin Streptomycin

Penicillin-streptomycin is a combination of antibiotics used to limit bacterial contamination in laboratory cell cultures, including models used in cancer research. Penicillin disrupts bacterial cell-wall synthesis, while streptomycin binds bacterial ribosomes and interferes with protein production, together inhibiting the growth of susceptible bacteria in culture media. Researchers commonly add this mixture when maintaining cancer cell lines, primary tumor cells, or other mammalian cultures for experiments such as drug screening, cellular analysis, and disease modeling. Its use helps preserve culture integrity and experimental consistency, although it complements rather than replaces careful aseptic technique.

This video demonstrates a technique for culturing bovine intervertebral discs in bioreactors to develop a three-dimensional organ culture model. Specific nutrient-deprived media and degenerative pressure conditions are applied to the discs, mimicking the pro-inflammatory and degenerative microenvironment observed during early-stage intervertebral disc disease. The disc heights are measured post-treatment to analyze the effects of nutrient deprivation and degenerative loading.
