Tumor Fraction Estimation

Tumor fraction estimation is the process of determining the proportion of tumor cells within a tissue or biological sample, a key measurement for interpreting cancer-related findings. It combines features such as tissue morphology, cellularity, and molecular signals to distinguish malignant material from normal cells; in genomic analyses, tumor fraction influences observed variant allele frequencies and helps estimate sample purity. Accurate estimates support variant detection, biomarker assessment, and comparison of tumor samples in cancer research. They also improve the interpretation of sequencing data and guide the selection of samples for molecular profiling, particularly when tumor and normal tissues are mixed.

This video describes a colorimetric assay for quantitatively measuring pyruvate in the cellular fractions of Caenorhabditis elegans. Utilizing the hydrogen peroxide produced from the enzymatic reaction of pyruvate oxidase with cellular pyruvate, a chromogenic dye is oxidized to produce a colored product, measured using colorimetry to determine the pyruvate concentration.

Here, we present a stereological method, the optical fractionator, used to quantify the formation of new neurons, and their survival, in the rat hippocampus following electroconvulsive stimulation. When correctly implemented, the sensitivity and efficiency of stereological methods ensures accurate estimates with a fixed and predetermined precision.
