Double Helix Structure

The double helix structure is the twisted, ladder-like arrangement formed by two complementary DNA strands, a molecular architecture that explains how genetic information is stored and copied. In DNA, a sugar-phosphate backbone supports inward-facing bases, whose specific hydrogen bonding, adenine with thymine and cytosine with guanine, holds the antiparallel strands together while allowing them to separate during replication. This structure links molecular geometry, noncovalent interactions, and sequence complementarity, making it central to chemistry and biochemistry. Understanding the double helix supports research on DNA synthesis, mutation, molecular recognition, biotechnology, and the design of nucleic-acid-based diagnostic and therapeutic tools.

This article describes a method to obtain a three-dimensional (3D) structure of helically assembled molecules using cryo-electron microscopy. In this protocol, we use HIV-1 capsid assemblies to illustrate the detailed 3D reconstruction procedure for achieving a density map by the iterative helical real-space reconstruction method.
