Nitrogen-15 Nmr

Nitrogen-15 NMR spectroscopy is an analytical technique that uses the nitrogen-15 isotope to reveal the chemical environments and bonding of nitrogen-containing molecules. In a strong magnetic field, nitrogen-15 nuclei align according to their spin states and absorb radiofrequency energy at characteristic resonance frequencies; chemical shifts and spin-spin coupling then provide structural information. Because nitrogen-15 has low natural abundance and sensitivity, researchers often use isotopically enriched samples or specialized acquisition methods. The technique helps characterize amines, amides, nitriles, heterocycles, proteins, and other compounds, supporting molecular structure determination, reaction monitoring, and studies of nitrogen-containing materials and biological systems.

Many organic, inorganic, and biological molecules contain spin-half nuclei such as nitrogen-15, fluorine-19, and phosphorus-31. As a result, NMR studies of these nuclei have found extensive applications in chemical and biological research. While fluorine-19 and phosphorous-31 have high natural abundances (100%) and positive gyromagnetic ratios, nitrogen-15 has a low natural abundance and a negative gyromagnetic ratio. However, nitrogen-15 is still preferred over nitrogen-14 (which has a high...

Nuclear magnetic resonance (NMR) spectroscopy can characterize structural protein dynamics in a residue-specific manner. We provide a hands-on protocol for recording NMR 15N R1 and R2 relaxation and {1H}-15N heteronuclear Overhauser effect (hetNOE) experiments, sensitive to the picoseconds to nanoseconds timescale.

The Nitrogen Cycle

2019

Nitrogen atoms, present in all proteins and DNA, are recycled between abiotic and biotic components of the ecosystem. However, the primary form of nitrogen on Earth is nitrogen gas, which cannot be used by most animals and plants. Thus, nitrogen gas must first be converted into a usable form by nitrogen-fixing bacteria before it can be cycled through other living organisms. The use of nitrogen-containing fertilizers and animal waste products in human agriculture has greatly influenced the...

Source: Laboratory of Dr. Henrik Sundén – Chalmers University of Technology Nuclear magnetic resonance (NMR) spectroscopy is a vital analysis technique for organic chemists. With the help of NMR, the work in the organic lab has been facilitated tremendously. Not only can it provide information about the structure of a molecule but also determine the content and purity of a sample. Compared with other commonly encountered techniques for organic chemists — such as thermal analysis and mass...
