Silver Mediated Glycosylation

Silver-mediated glycosylation is a chemical method for constructing glycosidic bonds by using silver reagents to activate glycosyl donors, making it important for assembling carbohydrates and glycoconjugates. In a typical reaction, a silver salt interacts with a leaving group such as a glycosyl halide, promotes formation of a reactive glycosyl species, and enables nucleophilic attack by an alcohol or other acceptor to form a new C–O bond. This approach supports the synthesis of oligosaccharides, glycosides, and biologically relevant carbohydrate structures, while donor design and reaction conditions influence yield and stereochemical outcome.

Gold Nanostar Synthesis with a Silver Seed Mediated Growth Method

We synthesized star shaped gold nanostars using a silver seed mediated growth method. The diameter of the nanostars ranges from 200 to 300 nm and the number of tips vary from 7 to 10. The nanoparticles have a broad surface plasmon resonance mode centered in the near infrared.

Analysis of SCAP N-glycosylation and Trafficking in Human Cells

We describe a modified method for membrane fraction isolation from human cells and sample preparation for the detection of SCAP N-glycosylation and total protein by using western blot. We further introduce a GFP-labeling method to monitor SCAP trafficking using confocal microscopy. This protocol can be used in regular biology laboratories.

Identification and Characterization of Protein Glycosylation using Specific Endo- and Exoglycosidases

Using specific glycosidases to remove sugars from glycoproteins followed by SDS-PAGE is a valuable method to detect glycan modifications on protein samples and is a good choice for initial glycobiology studies. Changes following deglycosylation can be detected as shifts in gel mobility or by staining with glycan sensitive reagents.
