Tlc Purification

TLC purification is a small-scale separation method that isolates compounds from a mixture using thin-layer chromatography, making it useful for checking and recovering products during chemical synthesis. A sample is applied as a band to a silica-coated plate, and a solvent moves through the stationary phase by capillary action; compounds travel at different rates because their adsorption to silica and solubility in the mobile phase differ. After development, separated bands are located under ultraviolet light or with a stain, removed from the plate, and eluted from the silica. This approach supports reaction workups, product cleanup, and rapid isolation when only milligram quantities are available.

Composition of polar lipid extracts and the fatty acid composition of individual glycerolipids are determined in a simple and robust lipid profiling experiment. For this purpose, glycerolipids are isolated by thin layer chromatography and subjected to transmethylation of their acyl groups. Fatty acyl methylesters are quantified by gas-liquid chromatography.
