Zebrafish Keratocytes

Zebrafish keratocytes are specialized stromal cells in the cornea that maintain tissue organization and respond to injury, making them useful for studying cell behavior in living animals. After corneal damage, local signals trigger cytoskeletal remodeling, adhesion turnover, and directed migration, while keratocytes can change their matrix-producing activity and cellular phenotype during repair. Their accessibility to imaging and regenerative capacity support research on wound healing, extracellular matrix organization, cell migration, and tissue regeneration. In developmental biology, zebrafish keratocytes provide a tractable model for linking cellular mechanisms with corneal formation, maintenance, and repair.

Zebrafish keratocytes migrate in cell sheets from explants and provide an in vitro model for the study of the mechanisms of collective cell migration in the context of epithelial wound healing. These protocols detail an effective way to establish primary explant cultures for use in collective cell migration assays.

A method to conditionally knockdown a target protein’s expression in the adult zebrafish retina is described, which involves intravitreally injecting antisense morpholinos and electroporating them into the retina. The resulting protein is knocked down for several days, which allows testing the protein’s role in the regenerating or intact retina.

Renal injuries incurred from nephrotoxins, which include drugs ranging from antibiotics to chemotherapeutics, can result in complex disorders whose pathogenesis remains incompletely understood. This protocol demonstrates how zebrafish can be used for disease modeling of these conditions, which can be applied to the identification of renoprotective measures.

Zebrafish Reproduction and Development

2023

The zebrafish (Danio rerio) has become a popular model for studying genetics and developmental biology. The transparency of these animals at early developmental stages permits the direct visualization of tissue morphogenesis at the cellular level. Furthermore, zebrafish are amenable to genetic manipulation, allowing researchers to determine the effect of gene expression on the development of a vertebrate with a high degree of genetic similarity to humans. This video provides a brief overview of...
