Knowledge Base Construction

Knowledge base construction is the systematic process of collecting, organizing, and representing information so that people or software can retrieve and use it reliably. In engineering, the process typically involves defining a domain scope, extracting facts from technical documents or databases, modeling entities and relationships, and encoding them in a structured schema with validation and version control. Search indexes, ontologies, and rule-based or machine-learning methods can then support efficient retrieval and automated reasoning. Well-constructed knowledge bases improve design decisions, troubleshooting, system integration, and technical documentation, while providing a foundation for expert systems, intelligent assistants, and data-driven engineering workflows.

This article describes RUGGED (Retrieval Under Graph-Guided Explainable disease Distinction), which integrates Large Language Model (LLM) inference with Retrieval-Augmented Generation (RAG). It draws evidence from expert-curated biomedical knowledge bases and peer-reviewed biomedical publications to synthesize new knowledge from up-to-date information, identify explainable and actionable predictions, and pinpoint promising directions for hypothesis-driven investigations.
