Mouse Stem Cells

Mouse stem cells are self-renewing cells that can generate specialized cell types, making them important models for studying mammalian development, disease, and gene function. Embryonic stem cells, isolated from the inner cell mass of the blastocyst, maintain pluripotency under defined culture conditions and can differentiate when developmental signals change. Researchers modify these cells using gene targeting or CRISPR-based editing before introducing them into embryos to produce genetically altered mice. This approach links specific DNA sequences to observable traits and supports studies of gene regulation, inheritance, disease mechanisms, drug responses, and regenerative biology.

Mouse Epidermal Neural Crest Stem Cell (EPI-NCSC) Cultures

Here we show our method to isolate mouse epidermal neural crest stem cells (EPI-NCSC). Technique involves micro-dissecting whisker follicles, isolating the bulge and placeing it into tissue culture. EPI-NCSC start to emigrate from bulge explants onto the substratum within 3 - 4 days.

Derivation of Hematopoietic Stem Cells from Murine Embryonic Stem Cells

This protocol details the derivation of transplantable hematopoietic stem cells from mouse embryonic stem cells (ESC) and their subsequent injection into lethally irradiated recipient mice. Briefly, ESC are differentiated as embryoid bodies, which are then infected with retroviral HoxB4 and co-cultured with OP9 stromal cells and hematopoietic cytokines.

An optimized protocol for the isolation of stem cells from the mouse salivary gland is described. The method employs enzymatic and mechanical digestion, and permits isolation of salispheres containing cells with characteristics of stem cells.
