Mrna Half-life Analysis

mRNA half-life analysis measures how long a messenger RNA molecule persists in a cell before its abundance falls by half, providing a quantitative view of post-transcriptional gene regulation. Researchers track transcript levels over time after transcription is stopped or newly synthesized RNA is labeled, then model the resulting decay to estimate half-life and compare stability among genes or conditions. In genetics, these measurements reveal how sequence elements, RNA-binding proteins, and degradation pathways influence gene expression, helping explain cellular responses, regulatory variation, and disease-associated changes in RNA stability.

Half-life of a Reaction

2020

The half-life of a reaction (t1/2) is the time required for one-half of a given amount of reactant to be consumed. In each succeeding half-life, half of the remaining concentration of the reactant is consumed. For example, during the decomposition of hydrogen peroxide, during the first half-life (from 0.00 hours to 6.00 hours), the concentration of H2O2 decreases from 1.000 M to 0.500 M. During the second half-life (from 6.00 hours to 12.00 hours), the concentration decreases from 0.500 M to...

Regulated mRNA Transport

2020

In eukaryotes, transcription and translation are compartmentalized; an mRNA is first synthesized in the nucleus and then selectively transported to the cytoplasm for protein synthesis. Before transport, a pre-mRNA undergoes several steps of post-transcriptional modifications including splicing, 5' capping, and the addition of a poly-adenine tail. Various proteins bind to the pre-mRNA during these modifications. The mRNA transport takes place with the help of multiple proteins playing specific...
