Autoimmune Rat Model

An autoimmune rat model is a laboratory rat that develops immune responses against the body’s own tissues, providing a controlled system for studying loss of self-tolerance and autoimmune disease. Autoimmunity may arise spontaneously or after researchers induce it by exposing the immune system to disease-associated antigens, triggering autoreactive T cells, autoantibodies, and tissue inflammation. In immunology and infection research, these models help clarify how immune dysregulation develops, distinguish autoimmune inflammation from pathogen-driven responses, and evaluate potential biomarkers or therapies. Their physiological complexity can also reveal treatment effects and mechanisms that simplified cell-based systems may not capture.

This report describes a method to induce chronic experimental autoimmune dry eye in Lewis rats through immunization with an emulsion of rat lacrimal gland extract, ovalbumin, and complete Freund's adjuvant, followed by the injection of lacrimal gland extract and ovalbumin into the forniceal subconjunctiva and lacrimal glands six weeks later.

Infection of mice with an Adenovirus expressing the major human autoantigen cytochrome P450 2D6 (hCYP2D6) recognized by sera of patients suffering from type 2 autoimmune hepatitis results in a persistent form of autoimmune-mediated liver disease characterized by extensive hepatitis, fibrosis and generation of a CYP2D6-specific immune response.
