Baseline Signal Recording

Baseline signal recording is the measurement of a system’s initial signal before an experimental stimulus, treatment, or infection, providing a reference for interpreting subsequent changes. In immunology and infection studies, researchers record the signal under defined resting conditions, then compare later measurements with the baseline to distinguish stimulus-associated responses from background variation, instrument drift, or sample-specific differences. This approach supports quantitative analysis of immune activation, pathogen detection, and host–pathogen interactions across assays that monitor cellular, molecular, optical, or electrical signals. Reliable baseline measurements improve experimental controls, strengthen comparisons between samples, and help identify subtle biological responses.

We present a method for collecting electrocorticographic signals for research purposes from humans who are undergoing invasive epilepsy monitoring. We show how to use the BCI2000 software platform for data collection, signal processing and stimulus presentation. Specifically, we demonstrate SIGFRIED, a BCI2000-based tool for real-time functional brain mapping.

Real-time In Vivo Recording of Arabidopsis Calcium Signals During Insect Feeding Using a Fluorescent Biosensor

This protocol outlines a simple method for analyzing calcium signals in plants generated by feeding hemipteran insects, such as aphids. Arabidopsis thaliana transformed with the GFP calcium biosensor GCaMP3 allow for the real-time in vivo imaging of calcium dynamics with a high temporal and spatial resolution.
