Human Tonsil Processing

Human tonsil processing is the laboratory preparation of surgically collected tonsil tissue for analyzing its immune cells and local infection responses. During processing, tissue is handled under sterile conditions, trimmed and mechanically dissociated, often with enzymatic digestion, then filtered and centrifuged to produce a viable single-cell suspension or defined tissue fractions. This workflow enables access to lymphocytes, antigen-presenting cells, epithelial cells, and microbial material for flow cytometry, cell culture, molecular profiling, and pathogen research. Standardized processing improves reproducibility when comparing healthy and diseased samples and supports studies of mucosal immunity, immune-cell activation, and host-pathogen interactions.

Palatine tonsils are a rich source of B and T lymphocytes. Here we provide an easy, efficient and rapid protocol to isolate B and T lymphocytes from human palatine tonsils. The method described has been specifically adapted for studies of the viral etiology of tonsil inflammation known as tonsillitis.
