Immune Cell Enumeration

Immune cell enumeration is the measurement of immune cell numbers, proportions, or concentrations in a biological sample, providing a quantitative view of immune status during health, infection, and disease. In flow cytometry-based enumeration, cells are labeled with fluorescent antibodies against lineage and activation markers, separated by light-scattering and fluorescence signals, and counted within defined analysis gates, often with counting beads or a known sample volume. These measurements help characterize leukocyte populations, track changes during infection, evaluate immune responses, and assess treatment effects. Standardized enumeration also supports diagnostics, vaccine studies, and comparisons across experiments.

A CD4 enumeration method, the α-test, is described which uses whole saliva to provide rapid and accurate CD4 counts.
