Late Stage Parasites

Late-stage parasites are parasites that have progressed to advanced developmental stages within a host or life cycle, a phase that often determines transmission, tissue damage, and disease persistence. As they mature, they may change surface antigens, occupy distinct tissues, acquire nutrients from the host, and modulate or evade immune responses, while host defenses involve antibody production, cellular immunity, and inflammatory signaling. Studying these stages helps researchers understand chronic infection, parasite development, and host–pathogen interactions, and supports the identification of diagnostic markers, therapeutic targets, and strategies for interrupting transmission in immunology and infection research.

This manuscript describes the use of a bioluminescent strain of African trypanosomes to enable the tracking of late stage infection and demonstrates how in vivo live imaging can be used to visualize infections within the central nervous system in real-time.

Separation of Plasmodium falciparum Late Stage-infected Erythrocytes by Magnetic Means

The paramagnetic properties of hemozoin are used to isolate late stages of Plasmodium falciparum-infected red blood cells growing in culture. The method is simple and fast and does not affect the subsequent invasive capabilities of the parasites.

Phenotypic Analysis of Rodent Malaria Parasite Asexual and Sexual Blood Stages and Mosquito Stages

Due to the striking similarities of the life cycle and biology of rodent malaria parasites to human malaria parasites, rodent malaria models have become indispensable for malaria research. Herein, we standardized some of the most important techniques used in the phenotypic analysis of wild-type and transgenic rodent malaria species.

Stage-specific isolation of mid-to-late Drosophila follicles is useful for a variety of purposes. Such follicles develop in culture, which allows for genetic and/or pharmacologic manipulations to be coupled with in vitro development assays and live imaging. Additionally, follicles can be used for molecular studies, such as isolating mRNA and protein.
