Particle Uptake

Particle uptake is the cellular process by which cells internalize solid or particulate material from their surroundings, an important event in immune defense and infection biology. In immune cells such as macrophages and dendritic cells, surface receptors recognize particle-associated signals and trigger actin remodeling, membrane engulfment, and formation of an intracellular vesicle that can mature into a degradative compartment. Uptake influences pathogen clearance, inflammatory signaling, and the processing of microbial material for antigen presentation. Measuring how cells capture fluorescent beads, nanoparticles, or microbial particles helps researchers compare immune-cell function, investigate host-pathogen interactions, and evaluate targeted delivery systems.

Measuring Microglial Uptake of Intravitreally-Delivered Fluorescent Particles Using Flow Cytometry

2025

This video demonstrates flow cytometry for measuring microglial uptake of fluorescent bioparticles after intravitreal delivery in a mouse eye. Flow cytometry detects live cells expressing CD11b and distinguishes phagocytic microglia cells exhibiting green fluorescence from other myeloid cells, including Ly6C-expressing monocytes and Ly6G-expressing neutrophils.

Radiolabeled Amino Acid Uptake Assay to Quantify Cellular Uptake of Amino Acids

2025

This video demonstrates an in vitro technique for estimating amino acid uptake by bone marrow-derived stromal cells. Na+-dependent amino acid transporters mediate the uptake of radiolabeled amino acids inside the cells, and the cellular uptake is estimated by measuring the radioactivity.

Mitochondrial Calcium Uptake Assay: A Plate Reader-Based Method for Measuring Calcium Uptake in Isolated Mitochondria

2025

This video describes a plate reader-based assay to measure mitochondrial calcium uptake in isolated mitochondria. The assay analyzes the fluorescence signal of a calcium-sensitive fluorescent dye to study the kinetics of mitochondrial calcium uptake and calcium overload.

Competence-Induced DNA Uptake in Staphylococcus aureus

Source: Cafini, F., et.al. Methodology for the Study of Horizontal Gene Transfer in Staphylococcus aureus. J. Vis. Exp. (2017)This video demonstrates competence-induced DNA uptake in Staphylococcus aureus through natural transformation. A chloramphenicol-sensitive strain engineered to constitutively express a sigma factor is cultured under nutrient-limiting conditions to enhance competence, then incubated with donor DNA carrying a resistance gene. Successful gene transfer is confirmed by the...

Subatomic Particles

2020

Dalton was only partially correct about the particles that make up matter. All matter is composed of atoms, and atoms are composed of three smaller subatomic particles: protons, neutrons, and electrons. These three particles account for the mass and the charge of an atom. The Discovery of the Electron The first clue about the subatomic structure came at the end of the 19th century when J.J. Thomson discovered the electron using a cathode ray tube. This apparatus consisted of a sealed glass...
