Tcr Transgenic Models

TCR transgenic models are experimental organisms, typically mice, engineered to express a predefined T cell receptor (TCR) that recognizes a specific peptide–MHC complex, enabling controlled study of antigen-specific immunity. Introducing TCR genes into hematopoietic cells produces a relatively uniform population of T cells with shared antigen recognition, while thymic selection and subsequent activation shape their development and function. In immunology and infection research, these models help trace T cell responses, measure clonal expansion, analyze tolerance and memory formation, and evaluate how pathogens or vaccines influence cellular immunity. They also support mechanistic studies of antigen presentation and immune-mediated disease.

This manuscript describes how to conduct (single molecule) Förster Resonance Energy Transfer (FRET)- based assays to measure the binding dynamics between T-cell antigen receptor (TCR) and antigenic peptide-loaded MHC molecules as they occur within the immunological synapse of a T-cell in contact with a functionalized planar supported lipid bilayer.

The intensely studied nematode worm Caenorhabditis elegans can be transgenically engineered to express the human β-amyloid peptide (Aβ). Induced expression of Aβ in C. elegans muscle leads to a rapid, reproducible paralysis phenotype that can be used to monitor treatments that modulate Aβ toxicity.

Induction of Intestinal Inflammation by Adoptive Transfer of CBir1 TCR Transgenic CD4+ T Cells to Immunodeficient Mice

In this protocol, a gut microbiota antigen-specific T cell adoptive transfer colitis model is described. CD4+ T cells are isolated from CBir1 TCR transgenic mice. These are specific for an immunodominant gut microbiota antigen CBir1 flagellin, which is transferred into recipient Rag1-/- mice, leading to intestinal inflammation.

The compartmentalization of proteins either within the plasma membrane or into intracellular locations is one regulatory mechanism that can greatly influence signaling outcomes; hence, to understand signaling it is important to study the spatial and temporal behavior of the proteins involved. We describe here a TIRF microscopy based system to study signal transduction in T cells, but is broadly applicable.
