Thp1 Monocytic Cells

THP-1 monocytic cells are a human acute monocytic leukemia-derived cell line widely used as an in vitro model of innate immune function. In suspension, these cells display monocyte-like characteristics; exposure to differentiating agents such as phorbol 12-myristate 13-acetate (PMA) activates protein kinase C signaling, promoting adherence and a macrophage-like phenotype. Researchers use THP-1 cells to examine phagocytosis, cytokine production, inflammatory signaling, and host responses to bacterial or viral infection. Their consistent growth and ability to model monocyte-to-macrophage differentiation support mechanistic studies, pathogen interaction experiments, and preliminary evaluation of immunomodulatory treatments in immunology and infection research.

A parasite-rescue and transformation assay with THP1 cells infected in vitro with Leishmania donovani has been optimized for anti-leishmanial drug screening. The assay involves differentiation of THP1 cells, infection with promastigotes, treatment with test drugs, controlled lysis of the infected macrophages, rescue of amastigotes, transformation to promastigotes and monitoring promastigote growth and proliferation with a fluorometric assay.

Differentiation and Polarization of Monocyte-Derived Cells into Macrophage-Like Cells

In this video, we demonstrate a procedure to induce the differentiation and polarization of peripheral blood-derived monocytes into M1 or M2 macrophages. Incubation with granulocyte-macrophage colony-stimulating factor, lipopolysaccharide, and interferon‐gamma yields pro-inflammatory M1 macrophages, while incubation with macrophage colony-stimulating factor and interleukin-4 induces anti-inflammatory M2 macrophage production.

This study compares two different methods of human monocyte isolation for obtaining in vitro dendritic cells (DCs). Monocytes are selected by adherence or negatively enriched by magnetic separation. Monocyte yield and viability along with MDDC viability, proliferation and CD11c/CD14 surface marker expression will be compared between both methods.

This video demonstrates the generation of immature, mature, and tolerogenic dendritic cells, or DCs, from human monocytes. The incubation of monocytes with growth factors and cytokines leads to the production of immature DCs. In response to immune regulatory molecules, immature DCs differentiate into tolerogenic and mature DCs.

This video demonstrates a method to study the responses to Mycobacterium tuberculosis infection in human M1 and M2 polarized monocyte-derived macrophages. The use of flow cytometry enables the study of a variety of surface markers associated with infected and uninfected macrophage populations.
