Buprenorphine Analgesia

Buprenorphine analgesia is the use of buprenorphine, a semisynthetic opioid, to relieve acute or chronic pain through distinctive receptor pharmacology. It acts primarily as a high-affinity, partial agonist at μ-opioid receptors, producing analgesia, and also antagonizes κ-opioid receptors; its strong receptor binding can reduce the effects of other opioids, while its partial-agonist profile creates a ceiling effect for respiratory depression without eliminating risk. Transdermal, buccal, and injectable formulations support different pain-management needs, with dosing and monitoring tailored to the patient and indication. Understanding these properties informs opioid selection and care for patients who also receive buprenorphine for opioid use disorder.

Pain is critical to various clinical pathologies, provoking an urgent need for effective management. Pain, whether acute or chronic, is a complex neurochemical process. Its alleviation depends on the type, with nonopioid analgesics effective for mild to moderate pain, such as musculoskeletal or inflammatory pain, while neuropathic pain responds best to anticonvulsants, tricyclic antidepressants, or serotonin/norepinephrine reuptake inhibitors. For severe acute or chronic pain, opioids may be...

This article describes a method for stabilizing long bone fractures that is based on the application of modified Ilizarov external fixators 1-3. After application of the fixators and creation of the bone injury, healing can be assessed, distraction osteogenesis can be performed, or non-union or critical sized defect can be created and used to study therapeutic interventions.

The surgical procedure used to induce experimental myocardial infarction in mice begins with left thoracotomy between the third and the fourth ribs in order to visualize the anterior surface of the heart and left lung. The left coronary artery is ligated, the chest is closed and the mouse is allowed to recover spontaneously.
