Evans Blue Assay

The Evans Blue assay is a quantitative method for assessing vascular permeability and albumin leakage from the bloodstream into tissues. In practice, intravenously administered Evans Blue dye binds strongly to serum albumin; when endothelial barriers become compromised, the dye-albumin complex extravasates and can be recovered from tissue and measured spectrophotometrically. In medicine, this assay helps evaluate blood-brain barrier disruption, inflammation, edema, and vascular injury in experimental models. Quantifying dye accumulation provides an accessible endpoint for comparing disease severity or treatment effects, although interpretation depends on tissue processing, vascular blood content, and assay conditions.

In this study, we describe a straightforward method to perform Evans Blue Dye (EBD) analysis on zebrafish larvae. This technique is a powerful tool for the characterization of skeletal muscle integrity and delineation of zebrafish models of muscular dystrophy, and is a valuable method for the development of novel therapeutics.

This video demonstrates an assay to monitor gut acidification in Drosophila flies. After the ingestion of food supplemented with a pH indicator dye, the acidification of the intestinal lumen results in a change in the color of the dye. Visual inspection of the digestive tract of the flies to examine the color change provides evidence of acidification.
