Gabaergic Function

GABAergic function describes the activity of signaling pathways that use gamma-aminobutyric acid (GABA), the principal inhibitory neurotransmitter in the central nervous system, to regulate neuronal excitability. GABA acts through GABA-A receptors, which open ligand-gated chloride channels, and GABA-B receptors, which activate G protein-coupled pathways that increase potassium conductance and reduce calcium entry. By limiting excessive neural firing, GABAergic signaling helps coordinate brain activity and influences anxiety, sleep, movement, and seizure susceptibility. In medicine, drugs that enhance or mimic this signaling, including benzodiazepines, baclofen, and some antiseizure agents, support treatment of anxiety, muscle spasticity, epilepsy, and related neurological disorders.

Transplantation of human pluripotent stem cell-derived GABAergic neurons generated by neuronal programming could be a potential treatment approach for neurodevelopmental disorders. This protocol describes the generation and transplantation of human stem cell-derived GABAergic neuronal precursors into the brains of neonatal mice, allowing long-term investigation of grafted neurons and evaluation of their therapeutic potential.
