Lung Volume Measurement

Lung volume measurement is the assessment of the air a person’s lungs can contain, move, and retain during breathing, providing important information about respiratory function. Spirometry records airflow and exhaled volume during controlled breathing maneuvers, while body plethysmography estimates total lung capacity and residual volume by measuring pressure changes as a person breathes inside an enclosed chamber. These measurements help distinguish obstructive from restrictive lung diseases, evaluate unexplained breathlessness, monitor disease progression, and assess responses to treatment. They also support preoperative evaluation and physiological research by quantifying how effectively the lungs expand and empty.

Measurement of the Pressure-volume Curve in Mouse Lungs

Here we present a protocol to simply and reliably measure the lung pressure-volume curve in mice, showing that it is sufficiently sensitive to detect phenotypic parenchymal changes in two common lung pathologies, pulmonary fibrosis and emphysema. This metric provides a means to quantify the lung’s structural changes with developing pathology.
