Organ Uptake Analysis

Organ uptake analysis is a method for measuring how much of a drug, tracer, biomolecule, or other substance accumulates in specific organs, revealing its distribution throughout the body. The analysis typically involves administering a detectable compound, allowing it to circulate and interact with tissues, then quantifying organ-associated signal or concentration through imaging, tissue sampling, or analytical assays. In medicine, these measurements help characterize pharmacokinetics, evaluate targeted delivery, identify organs involved in clearance or toxicity, and assess treatment performance. Organ uptake profiles can therefore guide dose selection, improve diagnostic imaging, and support the development of safer, more effective therapies.

Measurement of Extracellular Fluorescent Glucose Analog Depletion: A Surrogate Measurement Technique to Assess Glucose Uptake in Mouse Organs Ex Vivo

In this video, we demonstrate an ex vivo technique to measure extracellular glucose analog depletion in harvested mouse organs. It involves incubating the mouse tissue in a solution containing fluorescently-labeled deoxyglucose, which is taken up by the cells. The glucose analog uptake can be indirectly determined by measuring the fluorescence of the incubated medium.

Synaptosomal dopamine uptake and high-performance liquid chromatography analysis represent experimental tools to investigate dopamine homeostasis in mice by assessing the function of the dopamine transporter and levels of dopamine in striatal tissue, respectively. Here we present protocols to measure dopamine tissue content and assess the functionality of the dopamine transporter.
