Single Cell Pcr

Single-cell PCR is a molecular technique that amplifies and analyzes genetic material from an individual cell, enabling researchers to detect cell-specific mutations, transcripts, or pathogens that may be obscured in bulk samples. The method typically involves isolating a single cell, lysing it to release nucleic acids, and using sequence-specific primers for polymerase chain reaction; RNA targets require reverse transcription before amplification. In medicine, single-cell PCR helps characterize tumor heterogeneity, identify rare circulating cells, investigate immune-cell responses, and support genetic or infectious-disease testing. Its ability to link molecular profiles with individual cells strengthens precision diagnosis and improves understanding of disease mechanisms.

This video demonstrates the utility of digital PCR in a chip-in-a-tube format for the detection of single nucleotide variations in DNA samples. A single PCR reaction is partitioned into chambers that act as independent PCR reactions, and the detection of fluorescence signals from the amplified targets in the chambers is used to compute the frequency of the variant allele in the sample.
