Spheroid Culture Generation

Spheroid culture generation is a three-dimensional cell-culture method that forms compact, tissue-like clusters from dispersed cells, providing a model that can better reflect cellular interactions than conventional two-dimensional cultures. Cells aggregate through cell-cell adhesion when maintained in nonadherent or low-attachment conditions, such as suspension, hanging-drop, or specialized microwell systems, and may develop internal gradients of oxygen, nutrients, and waste. In medicine, spheroids support disease modeling, cancer research, drug screening, and studies of tissue responses. Their three-dimensional organization can improve evaluation of treatment effects and help bridge findings from laboratory cultures to more complex biological systems.

Spheroid Generation from Cell Lines: A Three-Dimensional (3D) Cell Culture Method

2023

Spheroids are a type of 3D cell culture model used to more accurately represent a cell’s environment compared to 2D cell culture monolayer models. In the example protocol, we will see scientists generate spheroids from a colorectal cancer cell line and apply live-cell imaging to track the spheroids’ growth.

Three-Dimensional Culture Systems: Spheroids and Organoids

2026

Three-dimensional (3D) culture systems, including spheroids and organoids, support New Approach Methodology (NAM)-aligned research by providing more physiologically relevant models than conventional two-dimensional cell cultures. Because they more closely resemble the structure and cellular interactions of tissues and organs, these systems can provide physiologically relevant data before advancing to animal studies. Spheroids as Disease and Drug Screening Models Spheroids are generated when one...

Generation of a Three-Dimensional Spheroid Model of Tumor Cell Invasion

2025

Source: Guyon, J., et al. A 3D Spheroid Model for Glioblastoma. J. Vis. Exp. (2020)The video demonstrates the generation of a three-dimensional spheroid model of tumor cell invasion. Spheroids are formed from human brain tumor cells and embedded in a collagen matrix. The tumor cells at the spheroid periphery adopt an invasive phenotype, secreting proteases to degrade the collagen and advancing to invade the surrounding matrix.

Generation of Spheroids from Human Pluripotent Stem Cells

This video demonstrates the transformation of human pluripotent stem cells (hPSCs) into spherical brain organoids. By using specific inhibitors, the hPSCs are guided to differentiate towards a neuronal lineage, forming neuroectodermal colonies. Special growth factors are then introduced, which encourage the self-organization and proliferation of the colonies, ultimately leading to the formation of spherical brain organoids.

Splitting Spheroids for Sub-culturing and Shipping: A Procedure for Propagating and Transferring Spheroids from Small Bowel Neuroendocrine Tumor
