Zucker Diabetic Fatty Rat

The Zucker Diabetic Fatty (ZDF) rat is a genetic rodent model of obesity-associated type 2 diabetes used to study metabolic disease and its complications. A mutation in the leptin receptor disrupts satiety signaling, promoting hyperphagia, insulin resistance, and obesity; in susceptible animals, progressive pancreatic beta-cell dysfunction leads to persistent hyperglycemia. Researchers use ZDF rats to investigate glucose and lipid metabolism, diabetes progression, vascular and renal complications, and responses to antidiabetic treatments. Because the model reproduces several features of human metabolic syndrome, it supports preclinical evaluation of disease mechanisms and therapeutic strategies while highlighting the relationship between obesity, insulin resistance, and beta-cell failure.

Pregnancy leads to significant changes to the fatty acid composition of maternal tissues. Lipid profiles can be obtained via gas chromatography to allow identification and quantification of fatty acids in individual lipid classes among rats fed various high and low fat diets during pregnancy.

Rapid Determination of the Thermal Nociceptive Threshold in Diabetic Rats

Here, we describe a rapid reliable and simple procedure to determine the lowest temperature at which rats or mice show nocifensive behavior, i.e. the thermal nociceptive threshold (TNT). This method applies a slowly increasing thermal stimulus allowing precise and reproducible estimation of TNTs with minimum, if any, stress to the animals.
