Cytoarchitecture

Cytoarchitecture is the organization of cells within a tissue, including their types, size, density, arrangement, and layering; in neuroscience, it provides a structural basis for distinguishing brain regions. Researchers assess cytoarchitecture by examining neuronal and glial distributions, cellular morphology, laminar patterns, and tissue sections revealed through histological staining or molecular markers. These features help define cortical and subcortical boundaries, relate anatomy to connectivity and function, and compare healthy, developing, or diseased nervous systems. Cytoarchitectonic maps therefore support brain atlases, interpretation of neuroimaging data, and investigations of how cellular organization contributes to cognition, behavior, and neurological disorders.

Vibratome Sectioning for Enhanced Preservation of the Cytoarchitecture of the Mammalian Organ of Corti

A simple procedure of vibratome sectioning the organ of Corti, followed by immunohistochemistry and confocal microscopy is described. This procedure allows for improved preservation of the fine cytoarchitecture of the mammalian organ of Corti, and consequently allows for accurate quantification of cell types.

Organotypic hippocampal slice cultures (OHSC) represent an in vitro model that simulates the in vivo situation very well. This manuscript details the fabrication of micro-tissue engineered neural networks: three-dimensional micron-sized constructs comprised of long aligned axonal tracts spanning aggregated neuronal population(s) encased in a tubular hydrogel. These living scaffolds can serve as functional relays to reconstruct or modulate neural circuitry or as biofidelic test-beds mimicking gray-white matter neuroanatomy.
