Electrophysiological Measurements

Electrophysiological measurements are techniques for recording the electrical activity of cells, tissues, or organs, providing a direct view of how nervous systems generate and transmit signals. In neuroscience, electrodes detect voltage changes or current flow produced by ion movement across neuronal membranes; intracellular recordings measure activity within a cell, whereas extracellular methods capture signals from nearby neurons or larger populations. These measurements can reveal resting membrane potential, action potentials, synaptic responses, and coordinated brain activity through methods such as patch clamp, single-unit recording, and electroencephalography. They support research on neural circuits, sensory processing, behavior, neurological disorders, and the effects of drugs or stimulation.

Insect olfactory systems provide unique opportunities for recording odorant-induced responses in the forms of electroantennograms (EAG) and single sensillum recordings (SSR), which are summed responses from all odorant receptor neurons (ORNs) located on the antenna and from those housed in individual sensilla, respectively.

The assessment and treatment of pain in infants is difficult because infants cannot verbally report their experience. In this video we describe quantitative electrophysiological methods and analysis techniques that can be used to measure the response to noxious events from the infant nervous system.

Measuring the Electrophysiological Activity of Neuronal Networks Using Micro-Electrode Arrays

The video demonstrates how to use a microelectrode array (MEA) to record the electrophysiological activity of neurons derived from human induced pluripotent stem cells (hiPSCs). Electrophysiological activity from a multiwell MEA containing a neuron-astrocyte co-culture is recorded to detect action potentials from multiple neurons in the network, confirming the successful differentiation of hiPSCs into neurons.
