Long-term Electrophysiology

Long-term electrophysiology is the sustained recording of electrical activity from nervous system cells or circuits over extended periods, enabling researchers to examine neural function beyond brief experimental sessions. Electrodes detect voltage changes produced by neuronal activity, while amplification, digitization, and stable recording interfaces preserve these signals for repeated or continuous analysis. In neuroscience, this approach helps track changes in firing patterns, network dynamics, learning, sensory processing, and disease-related activity over time. It is especially valuable for studying neural plasticity and evaluating chronic neural interfaces, where recording stability and signal quality are essential for interpreting biological changes.

In Vitro Calcium Imaging in Short-Term and Long-Term Rat Hippocampal Cultures

Source: Calvo-Rodríguez, et al. Fluorescence and Bioluminescence Imaging of Subcellular Ca2+ in Aged Hippocampal Neurons. J. Vis. Exp. (2015). This video demonstrates the use of fluorescence imaging to measure cytosolic calcium levels in short-term and long-term rat hippocampal cultures. By comparing young and aged neurons, it highlights how increased L-type voltage-gated calcium channel density and prolonged activation in aged neurons result in calcium overloading, a hallmark of neuronal aging.
